Reverse transcription polymerase chain reaction (RT-PCR) testing using nasopharyngeal swabs is the most sensitive method and is considered the gold standard for detection of SARS-CoV-2 virus. Easy-to-use and rapid testing platforms that can perform RT-PCR and provide lab-quality results are highly desired. We will describe the adaptation of Baebies’ FINDER®, which is a highly versatile multi-assay platform designed for neonatal and pediatric diagnostics built upon digital microfluidics (DMF) technology, to FINDER 1.5® platform to perform RT-PCR testing. DMF refers to a sophisticated method of manipulation of sub microliter droplets on a disposable cartridge. All steps including magnetic bead handling, sample concentration, sample purification, and RT-PCR were performed on the cartridge in <17 minutes. Heaters and sensors were integrated into a disposable digital microfluidic cartridge to enable ultra-rapid amplification of target DNA sequences. FINDER 1.5 is a toaster-sized instrument with all RT-PCR reagents (master mix with polymerase, primers, and probes) pre-loaded in a dry format in the disposable, single use cartridge. The FINDER 1.5 system utilizes standard lab methods such as RT-PCR and magnetic bead sample preparation, is easy to use, and rapid.
